Design: "Whole animal samples (minus their digestive system and a single leg removed for a DNA sample) were individually homogenized in TriFast using a Minilys homogenizer with the Precellys ceramic kit 1.4/2.8 mm. After homogenization total RNA was extracted from the samples following the manufacturer's instructions. All total RNA samples were checked for purity and quality using a NanoDrop spectrophotometer (NanoDrop Products, Wilmington, Delaware) and a 2100 BioAnalyzer (Agilent Technologies, Santa Clara, California). To decrease ribosomal RNA contamination and isolate mRNA, an mRNA enrichment was performed using the Dynabeads mRNA Purification Kit. Directional, strand specific RNA libraries for Illumina sequencing were prepared using the NEXTflex Directional RNA Seq Kit (dUTP based). Libraries were quality checked using an Agilent High Sensitiv DNA Chip on the 2100 BioAnalyzer. After quality check, libraries were sent to the Max-Delbruck-Centrum (Berlin, Germany) for cluster generation and sequencing. Libraries were individually barcoded using NEXTflex RNA-Seq Barcodes from Bioo Scientific and sequenced on a. Illumina HiSeq 2000 to generate 100-bp paired end reads. "
